چکیده

Plant lectins, because they bind to specific carbohydrate moieties, can be used to investigate the glyco-components of the cell surface. When considering the distribution, number, or mobility of lectin-binding sites, a distinction must be made between a natural distribution and a redistribution induced by binding with multivalent ligands (2, 4, 5, 8, 14, 15). In some cell types the induced redistribution results in the phenomenon of "cap-ping." Taylor et al. (14), on the basis of immuno-fluorescent studies, introduced the term capping to refer to the progressive movement and clumping of receptor-ligand complexes (e.g. binding site-lectin complexes) to form an aggregate of these com

Ferritin-conjugated plant agglutinins as specific saccharide stains for electron microscopy: application to saccharides bound to cell membranes.

An electron microscopic stain for specific saccharides was prepared by the conjugation of ferritin to concanavalin A, a plant agglutinin that specifically binds to oligosaccharides containing terminal d-glucose, d-mannose, or sterically related sugar residues. A technique was developed to allow topological visualization of erythrocyte and other membranes by means of transmission electron micros...

Adhesion of Phytophthora palmivora zoospores: detection and ultrastructural visualization of concanavalin A-receptor sites appearing during encystment.

The binding of concanavalin A (Con A) to the cell surface of zoospores and cysts of Phytophthora palmivora was studied by radiometry (125I-Con A), ultraviolet microscopy (fluorescein-Con A) and electron microscopy peroxidase-diaminobenzidine technique). Zoospores were found to secrete during the early stages of encystment a Con A-binding material susceptible to trypsin digestion. This glycoprot...

Schistosomula of Schistosoma mansoni clear concanavalin A from their surface by sloughing

The lectin concanavalin A (Con A) was used as a model probe to study the behavior of molecules bound to the surface of recently transformed schistosomula of Schistosoma mansoni. Con A binding was saturable (150-180 pg/organism) and specifically competed by alpha-methyl mannoside. Both FITC-Con A and 125-I-Con A were lost from the surface of schistosomula with a halftime of 8-10 h in culture in ...

Behenic Acid Monolayer and Bilayer Assemblies- A Study of Concanavalin A (Con A) Adsorption and its Interaction with Dextran Using Surface Plasmon Resonance Spectroscopy and Microscopy

Deposition of behenic acid (B.A) mono and bilayers onto gold coated surfaces was performed by Langmuir-Blodget dip-casting technique. Surface Plasmon resonance (SPR) and ellipsometry methods were employed for investigation of the monolayer and bilayer films. The adsorption of the biologically important molecule Concanavalin A (Con A) from bulk solution to these monolayers and bilayers as we...
